I would never replace one of the standard characters with a new one, but if I had to (like if Hasbro execs held me at gunpoint) I would drop Mrs. White (make all the staff innocent) and replace her with Miss Peach (a rival young character to Scarlet, and going by MD, possible relation to the deceased).

Huh
If you think about it, the male/female ratio is off anyway, when you think of dinner. There should at least be as many female guests as male, though for a "proper" dinner, there should be an equal number of each gender, including host, so actually the original game is shy two women. Replace Mr. Green with a butler character and change Mrs. White out of her maid outfit and into something more guest-y ... and then the ratios come out just fine.
You'd have Boddy at the head, Peacock on his right, Mrs. White on his left, Plum on Peacock's right and Mustard on White's left, and then Miss Scarlet at the foot.
